To do so you would need to compile your own build from the source code and apply a patch to enable USB. There is a ready-made development platform on the Rockbox site:

http://www.rockbox.org/wiki/VMwareDevelopmentPlatform

A word of warning: Some people reported problems, slow file transfers and/or corrupted files with this preliminary USB driver. It's probably better to patiently wait for the developers to fix the remaining bugs.

AFAIK this is the correct patch:
http://www.rockbox.org/tracker/task/11664

And there is this documentation on how to work with patches:

any opinions on if that patch is correct anyone? would hate to remove rockbox off my mp3 player then reinstall it and uninstall and reinstall if that patch doesn't work.

It's the same patch that dfkt enables in the builds he issues. That would be a easy way to test.
